The study was conducted by researchers at the University of Pennsylvania, Duke University, and the National Institute of Allergy and Infectious Diseases (NIAID). Instead of using live viruses, the new vaccine uses mRNAs containing two key proteins encoding Zika virus. In general, after mRNAs enter the receptor, they are cleared by the recipient's immune system within a few minutes, but the mRNAs used in the new vaccine are modified and ignored by the immune system, allowing easy access to the cells, and like live virus vaccines. After several weeks, the cells are induced to produce their encoded viral proteins, which in turn produce an immune effect. Compared to live virus vaccines, the new vaccine does not cause serious side effects and does not cause infection.
The results of a mouse test showed that injection of 30 micrograms of vaccine - equivalent to a small fraction of a dose of vaccine - triggered a rapid immune response in mice and protected mice from Zika virus two weeks later, and this Protection is still effective 5 months after inoculation. The macaque test also showed the same effect, with 50 micrograms of vaccine providing effective protection to macaques after 5 weeks.
Virus neutralization tests in both trials showed that the new vaccine induced receptors to produce high levels of antibodies, and the single-dose viral neutralization effect was 25 times that of the standard vaccine. Its antiviral effect peaks after a few weeks of inoculation, and it still has sufficient protection afterwards, and the effect may last for many years. The reason for this strong and lasting protection is that the vaccine is highly stimulating to CD4 helper T cells, which is important for maintaining long-term antibody immunity.
Researchers say that vaccine development strategies based on mRNAs are promising. In addition to having a strong and long-lasting immune effect, mRNAs vaccines are easier to produce and cost less than traditional viral vaccines. They hope to start clinical trials within a year to a year and a half.
